Four Novel Pharmaceutical Cocrystals of Oxyresveratrol, Including a 2 : 3 Cocrystal with Betaine

Cocrystal engineering can alter the physicochemical properties of a drug and generate superior candidate for formulation design. Oxyresveratrol (ORV) exhibits poor solubility in aqueous environments, thereby resulting bioavailability. Extensive cocrystal screening ORV with 67 formers (coformers) bearing various functional groups was therefore conducted using grinding, liquid-assisted solvent evaporation, slurry methods. Six cocrystals (ORV betaine (BTN), L-proline (PRL), isonicotinamide, nicotinamide, urea, ethyl maltol) were found, including four novel cocrystals. Powder X-ray diffraction, low frequency Raman spectroscopy, thermal analysis revealed unique crystal forms all obtained samples. Conventional infrared data differentiated by presence or absence hydrogen bond interacting aromatic ring ORV. The structures then elucidated single-crystal diffraction. Two new consisting : BTN (2 3) PRL H2O (1 2 1) identified, their solved. We report cocrystalline solids improved solubilities cage-like structures.
